Clone 2D7 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects LRRC8D. It targets an epitope within 18 amino acids from the N-terminal, extracellular domain.

Quality Control Testing

Evaluated by Western Blotting in mouse brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LRRC8D in mouse brain tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LRRC8D in lysates from wildtype HeLa cells but, not in lysate from HeLa LRRC8 A/B/C/D/E knockout cells.

Western Blotting Analysis; A 1:500 dilution from a representative lot detected LRRC8D in lysates from MEG01 cells treated with sh-SCR-mCherry and sh-LRRC8D-mCherry. (Data provided by Dr. Tarek Mohamed, Washington University School of Medicine in St. Louis, USA.)

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected LRRC8D in human cerebral cortex tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound LRRC8D peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected LRRC8D in HeLa cells.

Volume-regulated anion channel subunit LRRC8D (UniProt: Q7L1W4; also known as Leucine-rich repeat-containing protein 5, Leucine-rich repeat-containing protein 8D, HsLRRC8D) is encoded by the LRRC8D (also known as LRRC5) gene (Gene ID: 55144) in human. It is a non-essential component of the volume-regulated anion channel (VRAC, also named VSOAC channel) required to maintain a constant cell volume in response to extracellular or intracellular osmotic changes. LRRC8D-containing VRAC is important for the translocation of neutral compounds like myo-inositol, taurine, -aminobutyric acid (GABA) and prolonged transport of positively charged lysine. LRRC8D is a multi-pass membrane protein consists of four transmembrane domains at the N terminal and 13 leucine-rich repeats (LRR) at the C terminal. It is expressed in various tissues, with notable levels in the brain, lungs, and immune cells. It oligomerizes with other LRRC8 proteins (LRRC8A, LRRC8B, LRRC8C and/or LRRC8E) to form a heterohexamer. In vivo, the subunit composition may depend primarily on expression levels, and heterooligomeric channels containing various proportions of the different LRRC8 proteins may coexist. Studies report LRRC8D inhibits transport of immunoreactive cyclic dinucleotide GMP-AMP (2′-3′-cGAMP), an immune messenger produced in response to DNA virus in the cytosol. Although, VRAC are primarily known for passive anion efflux, study found that VRAC can influx the antibiotic Blasticidin S, as well as the essential cancer therapy drugs cisplatin and carboplatin. LRRC8D has been implicated in the proliferation, migration, death, and multidrug resistance of cancer cells through their involvement in various signal pathways. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.(Ref.: Planells-Cases, R., et al. 外形

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
