Evaluated by Western Blotting in Daudi cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Transcription factor PU.1 (SPI1) in Daudi cell lysate.

Tested Applications

Affinity Binding Assay: The binding affinity of this antibody to PU.1-Spi1 peptide (100 g/ml) was at least one thousand-fold (1,000X) higher than that obtained with the non-specific control peptide.

Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated PU.1/Spi1 in Daudi cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PU.1/Spi1 in HL-60 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

General description

Transcription factor PU.1 (UniProt: P17947; also known as 31 kDa-transforming protein) is encoded by the SPI1 gene (Gene ID: 6688) in human. SPI1 gene has been mapped to human chromosome 11p11.2. It belongs to the E26 transformation specific (Ets) family of transcription factors that binds (via the ETS domain) onto the purine-rich DNA core sequence 5′-GAGGAA-3′, also known as the PU-box. It can also can bind to RNA and interfere with pre-mRNA splicing. PU.1 has a N-terminal transcriptional activation domain with acidic amino acid sub domains, a central proline, glutamic acid, serine, and threonine-rich (PEST) region and a C-terminal (ETS) domain. The protein is mainly expressed in cells of the hematopoietic lineage. PU.1 is reported to be essential for the differentiation of myeloid cells and serves as a transcription activator involved in macrophage proliferation. PU.1 is overexpressed in some B cell lymphoma, Hodgkin, and non- Hodgkin lymphoma. Along with other transcription factors, it plays a vital role in fetal hematopoiesis and controls cellular communication in the immune system. High expression of Spi1 leads to senescence in erythroid progenitors and is regarded as critical step to target leukemia. Deletions in the SPI1 gene is implicated in acute myeloid leukemia (AML). SPI1 gene inactivation results in fetal death. Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal half of human Transcription factor PU.1 (SPI1).

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
