General description
THO complex subunit 1 (UniProt: Q96FV9; also known as Nuclear matrix protein p84, p84N5, hTREX84) is encoded by the THOC1 (also known as HPR1) gene (Gene ID: 9984) in human. The gene THOC1 is mapped to human chromosome 18p11.32-p11.31. THOC1 is a component of THO, a RNP (ribonucleoprotein) complex and is also involved in formation of larger RNP complexes like TREX (transcription export). THO participates in transcriptional elongation, transcript 3 end formation and nuclear export. THOC1 interacts with various RNA-binding proteins and is critical for the integrity of mRNA during transport. It has been shown to associate with elongating RNA polymerase II, and UAP56 (spliceosome RNA helicase DDX39B). Structurally, THOC1 is composed of multiple protein domains (including Dock and Death domains) that facilitate interactions with RNA and other proteins like THOC2. An intact death domain is needed for its apoptotic function. THOC1 is ubiquitously expressed with notable levels in tissues such as the brain and liver, and it is regulated during cellular stress responses. Subcellularly, THOC1 is localized primarily in the nucleus but, it shuttles between nucleus and the cytoplasm. Nuclear localization is required for induction of apoptotic cell death. Two isoforms of THOC1 have been described that are produced by alternative splicing. Dysregulation of THOC1 has been implicated in several diseases, including cancer and neurodegenerative disorders, where altered mRNA processing and transport contribute to disease pathology. THOC1 is up-regulated in breast tumors and is correlated with metastatic state of the tumor progression. It is also linked with prostate cancer aggressiveness and on the other hand, THOC1 suppresses cell growth in lung cancer cells. It also interacts with tumor suppressor retinoblastoma protein and is involved in the apoptotic activities. Studies demonstrated that knockdown of THOC1 reduces the proliferation of hepatocellular carcinoma (HCC) and, positions it as a predictive biomarker and therapeutic target in HCC.
