5,6-dihydroxyindole-2-carboxylic acid oxidase (UniProt: P17643; also known as DHICA oxidase, Catalase B, Glycoprotein 75, Melanoma antigen gp75, Tyrosinase-related protein 1, TRP, TRP-1, TRP1, EC:1.14.18) is encoded by the TYRP1 (also known as CAS2, TYRP, TYRRP) gene (Gene ID: 7306) in human. The encoded protein is involved in melanin biosynthesis and is a tyrosinase-like glycoenzyme that oxidizes DHICA into indole-5,6-quinone-2-carboxylic acid in the pathway. TRP1/TYRP1 is critical for skin/eye/hair pigmentation. TRP1 is a single-pass type I membrane protein and is expressed in melanosomes and endosomes in pigment cells. It is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to produce the mature protein that contains a large Lumenal, melanosome domain (aa 25-477), a transmembrane domain (aa 478-501), and a cytoplasmic tail (aa 502-537). TRP1/TYRP1 is regulated by the microphthalmia-associated transcription factor (MITF). MITF is a master regulator of pigmentation and a target of β-catenin in Wnt/β-catenin signaling during melanocyte differentiation. TRP1/TYRP1 is also regulated by the microRNA miR-145, and this microRNA is being examined as a possible therapy for hyperpigmentation. Studies have shown that difficulties in producing tyrosinase-like glycoenzymes in pure form have hampered the understanding of their activity and, the effect of mutations that cause albinism and pigmentation disorders. Recent studies have highlighted the involvement of TRP-1 in various diseases, including melanoma, where altered expression levels correlate with tumor progression and patient prognosis. TYRP1 mRNA expression level is considered as a prognostic marker for melanoma and is particularly useful when prognostic pathology parameters at the primary lesion are lacking. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Quality Control Testing

Evaluated by Immunohistochemistry in human skin tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected TYRP1 in human skin tissue sections.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TYRP1 in SK-MEL-28 cells .

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TYRP1 in human retina tissue lysate.

Affinity Binding Assay: A representative lot of this antibody bound TYRP1 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 3L4 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects TYRP1. It targets an epitope within 17 amino acids from the C-terminal cytoplasmic domain.

外形

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

0.3 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
