WD repeat-containing protein 5 (UniProt: P61964; also known as BMP2-induced 3-kb gene protein) is encoded by the WDR5 (also known as BIG3) gene (Gene ID: 11091) in human. WD repeat domain 5 (WDR5) is a highly conserved protein characterized by its WD40 domain structure, which facilitates protein-protein interactions. This protein is a core component of the MLL/SET1 histone H3 lysine 4 (H3K4) methyltransferase complexes, playing a pivotal role in chromatin remodeling and transcriptional regulation. WDR5 is ubiquitously expressed across various tissues, with notable enrichment in the brain, liver, and kidney, and is predominantly localized in the nucleoplasm and chromatin-associated regions. WDR5 contributes to histone modification by positioning the N-terminus of histone H3 for efficient methylation at lysine 4, thereby influencing gene expression and cellular differentiation. It interacts with multiple proteins, including MYC, PAXBP1, and components of the ATAC complex, linking transcriptional machinery to chromatin remodeling pathways. Dysregulation of WDR5 has been implicated in oncogenesis, particularly in cancers such as leukemia and neuroblastoma, where its role in chromatin accessibility and transcriptional control supports tumor progression. Quality Control Testing
Evaluated by Western Blotting in MCF7 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected WDR5 in MCF7 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected WDR5 in human and rat spleen tissue lysate, and a 1:10,000 dilution in C2C12 cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound with WDR5 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected WDR5 in A431 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected WDR5 in human testis tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 3N4 is a ZooMAb® rabbit recombinant monoclonal antibody that detects WDR5. It targets an epitope within 16 amino acids from the N-terminal half.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
