Clone 2C11 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects IFT88. It targets an epitope within 23 amino acids from the C-terminal region.

应用

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IFT88 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected IFT88 in lysates from Jurkat cells and mouse testis tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected IFT88 in human testis tissue sections.

Immunofluorescence Analysis: A 1:00 dilution from a representative lot detected IFT88 in human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound IFT88 peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Intraflagellar transport protein 88 homolog (UniProt: Q13099; also known as Polaris, Recessive polycystic kidney disease protein Tg737 homolog, Tetratricopeptide repeat protein 10, TPR repeat protein 10) is encoded by the IFT88 (also known as TG737, TTC10 ) gene (Gene ID: 8100) in human. The primary cilium is a microtubule-based mechano and chemo-sensory organelle that coordinates an array of cellular pathways during development and in tissue homeostasis, including Hedgehog (Hh), PDGFR and Wnt signaling. IFT88 is primarily localized at the basal body of primary cilia and within the cilia. Primary cilium is assembled and maintained via a process termed intraflagellar transport (IFT) in mammalian cells during growth arrest. IFT88 is a subunit of the IFT particle complex B required for the assembly of functional IFT. It is widely expressed in various tissues, particularly heart, brain, liver, lung, kidney, skeletal muscle and pancreas. Three isoforms of IFT88 have been described that are produced by alternative splicing. IFT88 protein has 9 Tetratricopeptide repeats (TPR) which are functional in protein-protein interactions. Defects in primary cilium assembly lead to severe developmental diseases and disorders referred to as ciliopathies. Polycystic kidney disease (PKD) is one of the first diseases identified to be linked to dysfunctional primary cilia. PKD was originally identified in the Oak Ridge Polycystic Kidney (ORPK) mouse harboring mutated IFT88 gene (Ift88orpk or Ift88Tg737NRpw). Studies have shown that loss of function of IFT88/primary cilia does not promote thyroid cancer cell proliferation, migration, and invasion but, IFT88 may play a role in metabolic reprogramming in thyroid cancer cells. IFT88 is commonly used as a primary cilium marker. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. 外形

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
