Application

Quality Control Testing

Evaluated by Western Blotting in Human umbilical vein endothelial cells (HUVEC) lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected PLVAP in Human umbilical vein endothelial cells (HUVEC) lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PLVAP in lysates from mouse lung and rat and human kidney tissues.

Affinity Binding Assay: A representative lot of this antibody bound with PLVAP peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PLVAP in TF-1 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 3K18 is a Z ZooMAb® recombinant rabbit monoclonal antibody that specifically detects PLVAP. It targets an epitope within 18 amino acids from C-terminal extracellular domain.

General description

Plasmalemma vesicle-associated protein (UniProt: Q91VC4; also known as MECA-32 antigen, Plasmalemma vesicle protein 1, PV-1) is encoded by the Plvap (also known as Pv1) gene (Gene ID: 84094) in murine species. PLVAP/PV-1 is an endothelial-specific integral membrane glycoprotein involved in the biogenesis and regulation of stomatal diaphragms of caveolae, trans endothelial channels (TECs), vesiculo-vacuolar organelles (VVOs) and diaphragms of endothelial fenestrae. It forms the vital radial fibrils, a key structural component, for the stomatal diaphragm (SD) and fenestral diaphragm (FD). Studies have suggested that PLVAP dimers form fibrils of the diaphragms with microvascular permeability. PLVAP is a single-pass type II membrane protein and has a short cytoplasmic domain (aa 1-26), a transmembrane domain (aa 27-47), and a long extracellular domain (aa 48-438). PLVAP contains three coiled coil regions (aa 140-160; 189-224; and 281-383). PLVAP protein is predominantly expressed in lung, kidney, spleen, heart, muscle, eye, pancreas, thyroid, thymus, submaxillary gland, prostate, epididymis, uterus, and liver. In mouse embryo it is expressed at 7 days post coitum (dpc). Its expression in the brain is negatively correlated with the differentiation of the vasculature to form the blood brain barrier. Experimental knockout models have demonstrated that loss of PLVAP leads to significant vascular dysfunction and premature mortality, highlighting its vital role in endothelial homeostasis and potential as a therapeutic target. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the C-terminal extracellular domain of mouse PLVAP.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
