Clone 2L23 is a ZooMAb® rabbit recombinant monoclonal antibody that detects IFNGR1. It targets an epitope within 19 amino acids from the N-terminal extracellular domain.

Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IFNGR1 in HepG2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected IFNGR1 in A549 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound with IFNGR1 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected IFNGR1 in one million human peripheral blood mononuclear cells (PBMC).

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected IFNGR1 in human kidney tissue and human placenta tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

General description

Interferon gamma receptor 1 (UniProt: P15260; also known as CD119, IFN-gamma receptor 1, IFN-gamma-R1, CDw119, Interferon gamma receptor alpha-chain, IFN-gamma-R-alpha) is encoded by the IFNGR1 gene (Gene ID: 3459) in human. Interferon-gamma receptor 1 (IFNGR1) is a transmembrane protein that plays a pivotal role in mediating immune responses. It is composed of an extracellular domain that binds interferon-gamma (IFN- ), a single transmembrane domain, and a cytoplasmic domain responsible for signal transduction. IFNGR1 forms part of the IFN- receptor complex alongside IFNGR2, and upon ligand binding, it activates the Janus kinase (JAK)-signal transducer and activator of transcription 1 (STAT1) signaling pathway, leading to the transcription of interferon-stimulated genes. These genes are critical for enhancing immune effector functions such as macrophage activation and antimicrobial activity. IFNGR1 is ubiquitously expressed across various tissues, with particularly high levels in immune cells such as macrophages and T cells. It is localized to the plasma membrane and undergoes internalization and recycling, which are tightly regulated to modulate receptor sensitivity. IFNGR1 interacts with other proteins, including JAK1, STAT1, and regulatory molecules such as AP-2 , which influences its expression levels and signaling efficacy. Dysregulation of IFNGR1 has been implicated in several diseases, including Mendelian susceptibility to mycobacterial disease (MSMD), cancer immune evasion mechanisms, and neurodegenerative conditions such as nigrostriatal degeneration. Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal extracellular domain of human IFNGR1.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

0.3 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
