Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Flotillin-2 in HDLM-2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Flotillin-2 in mouse brain and a 1:1,000 dilution in rat testis tissue lysate.

Affinity Binding Assay: A representative lot of this antibody bound with Flotillin-2 peptide with at least ten million-fold (10,000,000X) higher affinity than with non-specific control peptide.

Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated Flotillin-2 in HDLM-2 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Flotillin-2 in HDLM-2 cells.

Clone 1L10 is a ZooMAb^® rabbit recombinant monoclonal antibody that detects Flotillin-2. It targets an epitope within 25 amino acids from the N-terminal half.

Flotillin-2 (UniProt: Q14254; also known as Epidermal surface antigen (ESA), Membrane component chromosome 17 surface marker 1) is encoded by the FLOT2 (also known as ESA1, M17S1) gene (Gene ID: 2319) in human. Flotillin-2 is a highly conserved monotopic integral membrane protein associated with lipid rafts in cellular membranes. Flotillin-2 consists of an N-terminal stomatin/prohibitin/ flotillin/HflC/K (SPFH) domain and a C-terminal hydrophobic region, which are integral to its functions within membrane microdomains. The protein is widely expressed across various tissues and plays critical roles in cellular processes such as signal transduction, endocytosis, membrane trafficking, and cytoskeletal organization. Flotillin-2 forms hetero-oligomeric complexes with flotillin-1, which together modulate receptor signaling and intracellular trafficking. Flotillin-2 acts as a scaffolding protein within caveolar membranes or caveolae-like structures, facilitating the clustering and signaling of growth factor receptors and other signaling molecules. It participates in diverse processes, including T-cell activation, insulin signaling, and epidermal cell adhesion. Involved in oncogenic signaling, where it contributes to tumor cell motility, invasion, and transformation. In addition, Flotillin-2 has been implicated in axonal growth and neuronal signaling, underscoring its role in neurophysiology. Flotillin-2 localizes predominantly to vesicle-like structures within the cytoplasm and cholesterol-rich lipid rafts in the plasma membrane. Its expression is regulated by various post-translational modifications, including phosphorylation and palmitoylation, which modulate its activity and interaction with signaling pathways. Dysregulation of FLOT2 expression and function has been linked to pathological conditions such as inflammatory bowel diseases, cancer progression, and metabolic disorders. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
