General description

Thioredoxin-dependent peroxide reductase, mitochondrial (UniProt: P20108; also known as Antioxidant protein 1, AOP-1, PRX III, Perioredoxin-3, Protein MER5, Thioredoxin-dependent peroxiredoxin 3, EC:1.11.1.24) is encoded by the Prdx3 (also known as Aop1, Mer5) gene (Gene ID: 11757) in murine. The human and mouse genes are highly conserved, and they map to the regions syntenic between mouse and human chromosomes. Sequence comparisons with recently cloned mammalian homologues suggest that these genes consist of a family that is responsible for regulation of cellular proliferation, differentiation, and antioxidant functions. It is a housekeeping-type gene preferentially expressed in murine erythroleukemia (MEL) cells. PRDX3 is a mitochondrial antioxidant enzyme that plays a critical role in protecting cells from oxidative stress by catalyzing the reduction of hydrogen peroxide and other peroxides. Structurally, PRDX3 is characterized by a conserved thioredoxin-like fold, which contains a peroxidatic cysteine residue that forms a sulfenic acid intermediate during the reduction of peroxides. Two transcript variants encoding two different isoforms have been found for this gene. Dysregulation of PRDX3 expression has been implicated in various diseases, including neurodegenerative disorders and cancer, where it may influence tumorigenesis through modulation of cell proliferation and apoptosis pathways. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Quality Control Testing
Evaluated by Western Blotting in C2C12 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PRDX3 in C2C12 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PRDX3 in lysates from human kidney tissue, and a 1:10,000 dilution from a representative lot detected PRDX3 in lysate from mouse and rat kidney tissue.
Affinity Binding Assay: A representative lot of this antibody bound PRDX3 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis (IHC(P)): A 1:100 dilution from a representative lot detected PRDX3 in mouse kidney tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PRDX3 in C2C12 cells.

Biochem/physiol Actions
Clone 2I14 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects PRDX3. It targets an epitope within 24 amino acids from C-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
