General description
ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Gamma-aminobutyric acid receptor-associated protein (UniProt: O95166; also known as GABA(A) receptor-associated protein, MM46) is encoded by the GABARAP (also known as FLC3B, HT004) gene (Gene ID: 11337) in human. Gamma-aminobutyric acid receptor-associated protein (GABARAP) is a multifunctional protein belonging to the ATG8 family, which plays a key role in autophagy and receptor trafficking. GABARAP is a ubiquitin-like protein that undergoes lipidation to phosphatidylethanolamine (PE) on autophagosomal membranes, facilitating the formation of autophagosomes and selective cargo sequestration. Its crystal structure reveals interactions essential for membrane remodeling and cargo recruitment during autophagy. GABARAP contributes to GABA receptor trafficking and stabilization in synapses, which is critical for inhibitory neurotransmission in the central nervous system. It binds directly to the 2 subunit of GABA receptors, promoting their transport to the plasma membrane and preventing endocytosis. This stabilization of GABA receptors is associated with the modulation of synaptic currents and is clinically relevant for psychiatric disorders. Additionally, GABARAP interacts with autophagic receptors like p62 and NBR1 to mediate selective autophagy, targeting cellular organelles or aggregates for degradation. GABARAP has been implicated in cancer biology, particularly in suppressing epithelial-to-mesenchymal transition (EMT) and breast cancer progression via the AKT/mTOR signaling pathway. Western Blotting Analysis: A 1:10,000 dilution of this antibody detected GABARAP in C2C12 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected GABARAP in lysates from HeLa cells, and mouse and rat kidney tissues.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GABARAP in HeLa cells treated with 50 M chloroquine overnight, but not in untreated cells.
Affinity Binding Assay: A representative lot of this antibody bound with GABARAP peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions
Clone 2L12 is a ZooMAb® rabbit recombinant monoclonal antibody that detects GABARAP. It targets an epitope within 16 amino acids from the N-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
