General description

Intercellular adhesion molecule 2 (UniProt: P35330; also known as ICAM-2, Lymphocyte function-associated AG-1 counter-receptor, CD102) is encoded by the Icam2 (also known as Icam-2) gene (Gene ID: 15896) in mouse. ICAM-2/CD102 is a cell surface glycoprotein that is a part of the immunoglobulin superfamily. ICAM-2 is thought to act as a go-between during cell-cell adhesion interactions which are obligatory for immune system function. It is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 23-222), a transmembrane domain (aa 223-247), and a cytoplasmic domain (aa 248-277). The extracellular region contains two Ig-like C2-type 1&2 domains. ICAM-2 is expressed mainly in vascular endothelial cells and follicular dendritic cells, in lower amount on lymphocytes, monocytes and platelets. High expression levels are found in lung. This protein may play a role in lymphocyte recirculation by blocking LFA-1-dependent cell adhesion. It mediates adhesive interactions important for antigen-specific immune response, NK-cell mediated clearance, lymphocyte recirculation, and other cellular interactions important for immune response and surveillance. Dysregulation of ICAM-2 has been implicated in various pathological conditions, including chronic inflammatory diseases, cancer metastasis, and autoimmune disorders, where its altered expression can contribute to aberrant leukocyte infiltration and tissue damage. Studies have highlighted the potential of targeting ICAM-2 in therapeutic strategies aimed at modulating immune responses and treating inflammatory diseases. ICAM-2 may be a useful therapeutic target for oral squamous cell carcinoma. A recent study suggested ICAM2 to be a potential prognostic marker and a therapeutic target for gastric cancer. Application

Quality Control Testing

Evaluated by Flow Cytometry in BALB/c mouse splenocytes.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected mouse CD102 in one million BALB/c mouse splenocytes.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 3C4(mIC2/4) is a ZooMAb® recombinant rat monoclonal antibody that specifically detects CD102.

Physical form

Purified recombinant rat monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
