General description

A purified peptide (TASK) corresponding to amino acid residues 252-269 in the cytoplasmic C-terminus of human TASK1 (Accession O14649).

Application

Western blot: 1:200 using ECL on rat brain membranes.

Immunohistochemistry on rat brain sections.

Dilutions should be made using a carrier protein such as BSA (1-3%)

Optimal working dilutions must be determined by the end user.

Biochem/physiol Actions

The epitope is specific for TASK1 and is not present in any other known proteins tested so far.

SPECIES REACTIVITIES: The epitope is also highly conserved between mouse and rat (17/18 residues identical) but other species have not been tested.

Physical form

Affinity purified immunoglobulin. Lyophilized from phosphate buffered saline, pH 7.4, containing 1% BSA, 5% sucrose as a stabilizer and 0.025% sodium azide as a preservative. Reconstitute with 200 μL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).

Preparation Note

Maintain lyophilized material at -20°C for up to 12 months. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Other Notes

CONTROL ANTIGEN: Included free of charge with the antibody is 40 μg of control antigen (lyophilized powder). The stock solution of the antigen can be made up using 100 μL of sterile deionized water. For negative control, preincubate 1 μg of purified peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.
FUNCTION: pH-dependent, voltage-insensitive, background potassium channel protein. Rectification direction results from potassium ion concentration on either side of the membrane. Acts as an outward rectifier when external potassium concentration is low. When external potassium concentration is high, current is inward.

SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein (Potential).
TISSUE SPECIFICITY: Widespread expression in adult. Strongest expression in pancreas and placenta. Lower expression in brain, lung, prostate, heart, kidney, uterus, small intestine and colon.
MISCELLANEOUS: Inhibited by external acidification. Activated by halothane and isoflurane.
SIMILARITY: Belongs to the two pore domain potassium channel (TC 1.A.1.8) family.
