General description
T-cell surface glycoprotein YE1/48 (UniProt: P20937; also known as Lymphocyte antigen 49a, Ly-49a, T lymphocyte antigen A1) is encoded by the Klra1 (also known as Ly-49, Ly-49a, Ly49, Ly49A) gene in murine species. LY49A is a single-pass type II membrane protein that serves as an inhibitory receptor on natural killer (NK) cells for H-2d(Dd) MHC class I molecules on target cells. It inhibits the activity of NK cells thereby preventing cell lysis. Ly49A-mediated NK cell inhibition is shown to be dependent on a lateral binding site, which is located beneath the peptide-binding platform of Dd. Ly49A is reported to contribute to target cell adhesion and accumulate at synapses with Dd-expressing target cells when NK cells themselves lack Dd. In cases where NK cells display Dd expression Ly49A does not contribute to adhesion. Higher expression of LY49A is observed in T-lymphoma cell lines and normal lymphocytes exhibit low expression. LY49A has a cytoplasmic domain (aa 1-44), a transmembrane domain (aa 45-66), and an extracellular domain (aa 67-262). (Ref.: Back, J., et al. (2007). Proc. Natl. Acad. Sci. USA 104(10); 3978 3983).
