Biochem/physiol Actions

A 6-thioguanine resistant mutant of mouse L cell fibroblasts (Mouse strain C3H/An). A9 lacks guanylic acid-inosinic acid pyrophosphorylase and was isolated by virtue of resistance to 6-thioguanine (3 μg/ml). ECACC has two catalogue entries for the A9 cell line which differ in that they were deposited by different groups: A9 (Sigma Catalogue number 85011426) and A9(Hamprecht) (Sigma Catalogue number 08062526). Several other cell lines in the ECACC catalogue have been derived from A9(Hamprecht).
Mouse Fibroblast (L cells)

Preparation Note

DMEM + 2 mM glutamine + 10% Foetal Bovine Serum (FBS) + 0.1 mM 6-thioguanine
Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.05% trypsin/EDTA; 5% CO2; 37°C.
