WEHI 164 Cell Line from mouse

Tumourigenicity and cytotoxicity studies, chromium release assays

Established from a fibrosarcoma induced by subcutaneous injection of methylcholanthrene in BALB/c mice. Highly sensitive after pre-treatment with actinomycin D to human cytotoxic monocytes, to tumour necrosis factor and to lymphotoxin. It provides convenient target cells for short-term (51) chromium release assays.

Mouse BALB/c fibrosarcoma

DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).

Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm² using 0.25% trypsin or trypsin/EDTA; 5% CO₂; 37°C.
