Study of biology of pancreatic islet cells, synthesis, storage and secretion of insulin and somatostatin
Biochem/physiol Actions
Established from a transplantable pancreatic tumour of the islets of Langerhans. The tumour originated in an irradiated NEDH inhibited albino rat and was maintained by serial transplantation in Nedh rats. After transplantation into athymic nude mice the cell line RIN-m was established from the fourth nude mouse heterotransplant, 9 days after the tumour was removed cells were exposed to cystine-free medium resulting in the complete elimination of fibroblast-like cells. RIN-m cells secrete insulin and somatostatin. High levels of L-dopa-decarboxylase and membrane bound secretory granules were detected. RIN-5F (95090402) was established as a secondary clone from RIN-m.

Preparation Note
RPMI 1640 + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 1-3x10,000 cells/cm2 using 0.05% trypsin or trypsin/EDTA; 5% CO2; 37°C.
