Biochem/physiol Actions
RIN-5F is a secondary clone of the rat islet tumour cell line RIN-m (Sigma Catalogue number. 95071701). Cloning was performed by limited dilution in RPMI 1640, 10%FBS and 20% conditioned medium. The conditioned medium contained growth hormones and prolactin being produced by rat pituitary tumour cells GH3 (Sigma Catalogue number. 87012603). RIN-5F cells secrete insulin, produce high levels of the key APUD enzyme, L-dopa-decarboxylase and contain membrane-bound secretory granules.

Preparation Note
RPMI 1640 + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C. Cells grow in small islands and do not form a confluent monolayer.
