General description

10 mM Trizma® hydrochloride (pH 8.5) is a widely used buffer in molecular biology applications such as nucleic acid extraction and purification. No mixing or pH adjustment is necessary. Guaranteed accuracy for this buffer is ± 0.2 pH units at 25°C.

Application

Trizma® hydrochloride solution can be used:
  • As an elution buffer after PCR cleanup
  • As an elution buffer after DNA extraction

Preparation Note

Allow the buffer to reach room temperature before use.

Other Notes

Prepared with BioUltra Trizma Base for molecular biology in 18 megaohm UV irradiated water. pH-adjusted and 0.2 μm filtered. The buffer was tested by 35 cycles of PCR for prokaryotic and eukaryotic DNA contamination. To keep the buffer contamination-free, it is recommended to use gloves and barrier-tip pipettes for dispensing. Between uses, keep the bottle closed and refrigerated, away from areas where DNA samples and PCR products may be stored.
The pH values of all buffers are temperature and concentration dependent. For Tris buffers, pH increases about 0.03 unit per °C decrease in temperature, and decreases 0.03-0.05 unit per ten-fold dilution. For precise applications, use a carefully calibrated pH meter with a glass/calomel combination electrode.
