Specificity

Clone 2C16 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects human Pigment epithelium-derived factor (PEDF).

Immunogen

GST/His-tagged recombinant fragment corresponding to 152 amino acids from the C-terminal half of human Pigment epithelium-derived factor (PEDF).

Application

Affinity Binding Assay: A representative lot of this antibody bound human recombinant PEDF with a KD of 8.1 x 10-9 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PEDF (SERPIN F1) in human kidney tissue sections.

Anti-PEDF (SERPIN F1), clone 2C16 ZooMAb, Cat. No. ZRB1501, is a recombinant Rabbit monoclonal antibody that specifically targets PEDF and is tested for use in Affinity Binding Assay, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Pigment epithelium-derived factor (UniProt: P36955; also known as PEDF, Cell proliferation-inducing gene 35 protein, EPC-1, Serpin F1) is encoded by the SERPINF1 (also known as PEDF) gene (Gene ID: 5176) in human. Serpin F1 is a monomeric secreted protein that is synthesized with a signal peptide (aa 1-19) that is subsequently cleaved off to produce the mature form. Serpin F1 is synthesized by multiple cells, including retinal pigment epithelium, fibroblasts, mammary epithelium, adipocytes, hepatocytes and osteoblasts. It is enriched in retinal pigment epithelial cells and blood plasma. It displays neurotrophic activity and induces extensive neuronal differentiation in retinoblastoma cells. Its N-terminal (aa 44-121) displays neurite outgrowth-inducing activity while the C-terminal exposed loop (aa 382-418) is reported to be essential for its serpin activity. Serpin F1 also serves as a potent inhibitor of angiogenesis. Serpin F1 contains three beta sheets, 8 10 alpha helices, and a C terminal reactive center loop. In the cell, serpin F1 undergoes phosphorylation at Ser227 by protein kinase A that increases its anti-angiogenic activity and extracellular serpin F1 can be phosphorylated on serine 24 and serine 114 by casein kinase 2 that promotes its neurotrophic activity. Mutations in SERPINF1 gene are shown to cause Osteogenesis imperfecta 6 that is characterized by low bone mass, bone fragility and susceptibility to fractures after minimal trauma. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

0.03 mg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
