General description
Neuronal acetylcholine receptor subunit alpha-7 (UniProt: P36544; also known as nAChR7, Nicotinic acetylcholine receptor subunit alpha-7) is encoded by the CHRNA7 (also known as NACHRA7) gene (Gene ID: 1139) in human. CHRNA7 encodes the alpha-7 subunit of the neuronal nicotinic acetylcholine receptor (nAChR), which is part of a superfamily of ligand-gated ion channels. These receptors mediate fast synaptic signal transmission and play critical roles in neuronal communication, plasticity, and cognitive function. The alpha-7 subunit forms homopentameric receptors that are highly permeable to calcium ions and characterized by rapid desensitization. These receptors are activated by the neurotransmitter acetylcholine and other ligands, facilitating ion flux that influences neuronal excitability and signaling. CHRNA7 is ubiquitously expressed in both the central nervous system (CNS) and peripheral tissues. Within the CNS, these receptors are particularly abundant in regions involved in learning, memory, and sensory processing, such as the hippocampus and cortex. The alpha-7 nAChRs interact with various neurotransmitter systems and are integral to pathways such as Acetylcholine binding and downstream events, Regulating synaptic plasticity and excitatory neurotransmission. Mutations or deletions in CHRNA7 are linked to several neuropsychiatric and developmental disorders, including Chromosome 15q13.3 Deletion Syndrome which is associated with intellectual disability, epilepsy, and schizophrenia. Dysregulation of CHRNA7 signaling contributes to deficits in attention and memory. Application
Quality Control Testing
Evaluated by Western Blotting in IMR-32 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CHRNA7 in IMR-32 cell lysate.
Tested Applications
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CHRNA7 in human cerebral cortex tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound with CHRNA7 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1K16 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CHRNA7. It targets an epitope within 24 amino acids from the N-terminal extracellular domain.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
