General description

Heterogeneous nuclear ribonucleoproteins A2/B1 (UniProt: P22626; also known as hnRNP A2/B1) is encoded by the HNRNPA2B1 (also known as HNRPA2B1) gene (Gene ID: 3181) in human. HNRNPA2B1 plays a critical role in various aspects of RNA metabolism, including splicing, transport, stability, and translation. HNRNPA2B1 contains two RNA recognition motifs (RRMs) that facilitate binding to RNA sequences, enabling it to influence the processing and regulation of mRNA precursors and other RNA molecules. It is ubiquitously expressed in most tissues, with higher levels observed in the brain, where it is involved in neuronal RNA processing and regulation. HNRNPA2B1 is predominantly localized in the nucleus, where it associates with heterogeneous nuclear RNA (hnRNA) to form ribonucleoprotein complexes, although it can shuttle between the nucleus and the cytoplasm to execute its functions. Dysregulation of HNRNPA2B1 is implicated in several diseases, particularly neurodegenerative disorders such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). HNRNPA2B1 has also been linked to cancer; altered expression affecting tumor growth, metastasis, and response to therapy. Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal half of human HNRNPA2B1.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK-293 cell lysate.

Western Blotting Analysis: A 1:20,000 dilution of this antibody detected HNRNPA2B1 in HEK-293 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:20,000 dilution from a representative lot detected HNRNPA2B1 in K-562 cell lysate and a 1:10,000 dilution in NIH/3T3 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HNRNPA2B1 in HEK-293 cells.

Affinity Binding Assay: A representative lot of this antibody bound with HNRNPA2B1 peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected HNRNPA2B1 in human small intestine and rat testis tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1J11 is a ZooMAb® rabbit recombinant monoclonal antibody that detects HNRNPA2B1. It targets an epitope within 19 amino acids from the N-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
