General description

Insulin-like growth factor 2 (UniProt: P01344; also known as Insulin-like growth factor II, Somatomedin-A, T3M-11-derived growth factor, Insulin-like growth factor II Ala-25 Del, Preptin) is encoded by the IGF2 (also known as IGF-2; ORF: PP1446) gene (Gene ID: 3481) in human. IGF2 is a key growth factor, composed of a single polypeptide chain stabilized by disulfide bonds, sharing significant homology with insulin. IGF2 plays a pivotal role in fetal development, cell proliferation, and differentiation by binding to IGF receptors, including IGF1R and IGF2R, and insulin receptors. Its activity is modulated by IGF-binding proteins (IGFBPs), which regulate its bioavailability and receptor interactions. IGF2 is predominantly expressed during embryogenesis, with lower levels persisting in adult tissues such as the liver and brain. IGF2 resides in the extracellular matrix and circulates in plasma, where it interacts with IGFBPs to mediate endocrine and paracrine signaling. Dysregulation of IGF2 expression, often through epigenetic alterations, has been implicated in various diseases, including cancer, Beckwith-Wiedemann syndrome, and Silver-Russell syndrome, where aberrant IGF2 signaling contributes to tumorigenesis and growth abnormalities. IGF2 also participates in metabolic pathways, influencing glucose homeostasis and lipid metabolism. Its involvement in disease highlights its therapeutic potential, with strategies targeting IGF2 signaling pathways under investigation for cancer and metabolic disorders. Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal half of human IGF2.

Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 conditioned media.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected IGF2 in HepG2 conditioned media.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected a recombinant fragment of human IGF2.

Affinity Binding Assay: A representative lot of this antibody bound with IGF2 peptide with at least one billion-fold (1,000,000,000X) higher affinity than with non-specific control peptide.

Enzyme Immunoassay: A representative lot of this antibody detected IGF2 protein in ELISA application.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 2G15 is a ZooMAb® rabbit recombinant monoclonal antibody that detects IGF2. It targets an epitope within 22 amino acids from the N-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
