General description

Rho GDP-dissociation inhibitor 1 (UniProt: P52565; also known as Rho GDI 1, Rho-GDI alpha) is encoded by the ARHGDIA (also known as GDIA1) gene (Gene ID: 396) in human. This gene encodes a protein that plays a key role in the regulation of signaling through Rho GTPases. The encoded protein inhibits the disassociation of Rho family members from GDP (guanine diphosphate), thereby maintaining these factors in an inactive state in the cytoplasm. implicate RhoGDI1 in various processes involving the dynamic reorganization of cell-cell junctions. Activity of this protein is important in a variety of cellular processes, and expression of this gene may be altered in tumors. ARHGDIA is expressed in various tissues, with notable expression in the brain, heart, and skeletal muscle, reflecting its essential role in neuronal signaling and muscle function. It is predominantly localized in the cytoplasm and can translocate to the plasma membrane under specific conditions, influencing its regulatory capacity on Rho GTPases. Structurally, ARHGDIA contains a Rho-GDI domain that facilitates the binding and sequestration of Rho proteins such as RhoA, Rac1, and Cdc42 in their inactive GDP-bound form, preventing their activation and subsequent downstream signaling. Alternate splicing results in multiple transcript variants. Dysregulation of ARHGDIA has been implicated in several diseases, including neurological disorders and cancer, where altered Rho signaling contributes to tumor progression and metastasis. It is down-regulated in hepatocellular carcinoma (HCC). Absence of ARHGDIA helps in hepatic tumorigenesis. Mutations in this gene have been found in individuals with nephrotic syndrome, type 8. Recent studies implicate RhoGDI1 in various processes involving the dynamic reorganization of cell-cell junctions. Application

Quality Control Testing

Evaluated by Western Blotting in SK-OV-3 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected ARHGDIA in SK-OV-3 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected ARHGDIA in lysates from rat liver tissue and U-251 MG cells, and a 1:1,000 dilution from a representative lot detected ARHGDIA in mouse liver tissue lysate.

Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated ARHGDIA in SK-OV-3 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound ARHGDIA peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1K12 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects ARHGDIA. It targets an epitope within 21 amino acids from the N-terminal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
