General description

85 kDa
磷脂酰肌醇3激酶(PI3激酶)是一种脂质激酶,据信在生长信号通路中磷酸化第二信使。PI3K是催化110 kDa(p110)和调节85 kDa(p85、p50或p55)亚基的异源二聚体。p85单元由两个密切相关的蛋白质组成:与催化亚基紧密结合的p85 α和功能未知的p85 β。它们有62%的氨基酸序列相同。p85 α缺乏PI3K激酶活性,但可以作为PI3K和配体激活受体之间的纽带,成为催化亚基p110的调节剂。这两种亚型都与活化的受体结合,并作为酪氨酸激酶底物。

Analysis Note

对照
阳性抗原对照:货号12-301,未刺激的A431细胞裂解液。加入2.5µL 2-巯基乙醇/100µL裂解液,煮沸5分钟以减少制备量。在微凝胶每个泳道上样20µg经还原的裂解液。
通过蛋白质印迹法对人A431细胞、大鼠L6成肌细胞的RIPA裂解液进行常规评估。

蛋白质印迹分析:
0.5-2 µg/mL该批次在人A431细胞的RIPA裂解液中检测到PI3激酶;先前批次在大鼠L6成肌细胞的RIPA裂解液中检测到PI3激酶。
