General description
组蛋白H2A.X,也称H2A/X或H2A.X,由基因名称H2AFX和H2AX编码,是组蛋白H2A的变体,与基因组DNA相似。但是,组蛋白在结构上与H2A家族的其他成员不同,因为它具有一个C末端尾巴,该尾巴包含Ser139残基,该残基响应于双链DNA的断裂而被磷酸化。H2A.X的磷酸化是由ATM/ATR蛋白介导的快速过程。作为组蛋白H2A家族的成员,组蛋白H2A.x(H2A.x)是在核小体子集中替代常规H2A的变体组蛋白H2A。H2A.x参与核DNA上双链断裂(DSB)损伤的DNA修复。双链DNA断裂后,H2A.x在丝氨酸139处被ATM激酶迅速磷酸化,并变成γ-H2AFX。该磷酸化步骤可以从DSB的实际位点延伸至数千个核小体,并且可以标记周围的染色质,以募集DNA损伤信号传导和修复(1)所需的蛋白质。作为严重DNA损伤引起的细胞凋亡过程中翻译后修饰的一部分,磷酸化H2A.x的高表达被认为是细胞凋亡的准确指标。
