General description

p53肿瘤抑制基因整合了许多控制细胞生命和死亡的信号。最近发现了几种参与p53信号传导的新分子,包括Chk2,p53R2,p53AIP1,Noxa,PIDD和PID/MTA2。检查点激酶Chk2是酵母Cds1/Rad53的哺乳动物同源物。响应DNA损伤,检查点激酶ATM磷酸化并激活Chk2,而Chk2又直接磷酸化并激活p53。Chk2充当ATM下游效应子,以介导p53的激活。Chk2还磷酸化并激活BRCA1,BRCA1是在乳腺癌和卵巢癌中突变的肿瘤抑制基因的产物。
