一般描述

在CpG二核苷酸的情况下，NA甲基转移酶会甲基化胞嘧啶的5-位。DNA甲基化对于正常胚胎发育、印迹和X染色体失活至关重要。甲基CpG结合蛋白（MeCPs）可特异性识别DNA的甲基化区域，并直接或通过与已知共抑制蛋白（包括组蛋白脱乙酰基酶蛋白家族成员）结合来抑制转录。

免疫原

KLH偶联肽，对应于小鼠MeCP2的氨基酸465-478（C-PRPNREEPVDSRTP），同时N端添加了半胱氨酸以进行偶联。

应用

使用在WB中验证的抗MeCP2抗体（兔多克隆抗体）检测MeCP2（也被称为MeCP-2蛋白、智力缺陷伴x染色体16、智力缺陷伴x染色体79、甲基CpG结合蛋白2）。

可识别MeCP2，分子量75 kDa。 在HeLa核提取物中还观察到约75 kDa的蛋白质。 该蛋白通过将抗体与免疫原性肽进行预孵育而发生竞争，这表明某些HeLa菌株含有MeCP2或相关蛋白，这与先前的报道相反。
基于免疫原性序列的保守性，可预测其会与人发生交叉反应。
