Rosetta宿主菌株是BL21衍生物，旨在增强真核蛋白的表达，该蛋白含有在大肠杆菌中很少使用的密码子。这些菌株在兼容的氯霉素抗性质粒上为AGG、AGA、AUA、CUA、CCC、GGA密码子提供tRNA。因此，Rosetta菌株提供了“通用”翻译，否则该翻译受大肠杆菌密码子使用的限制。tRNA基因由其天然启动子驱动。在Rosetta（DE3）pLysS中，稀有tRNA基因存在于携带T7溶菌酶基因的相同质粒上。

DE3表明宿主是λDE3的溶原菌，因此在lacUV5启动子的控制下携带T7 RNA聚合酶基因的染色体拷贝。这样的菌株适用于通过用IPTG诱导从克隆在pET载体中的靶基因生产蛋白质。
