General description

铁调节蛋白(IRP-1 和 IRP-2)是胞质mRNA结合蛋白,通过调节铁蛋白、Tfrs以及位于5′-UTR的含铁响应性元件的其它蛋白,控制胞内铁水平。在铁饥饿时,IRP以高亲和力结合IRE。 IRP与铁蛋白 IRE的结合抑制其转录。但是,当细胞的铁充足时,IRP失去结合IRE的能力,使得铁蛋白高效翻译,并降低Tfr mRNA 的半衰期。尽管IRP1和 IRP-2 具有显著的蛋白序列同源性,它们的组织分布和调节模式却不同。IRP-2/IRE-BP2(大鼠/人963 aa)比IRP-1 的丰度较低,并缺乏乌头酸酶活性。它的N端具有独特的73-aa插入序列。在高水平铁的情况下, IRP-2被快速靶向,实现了蛋白酶体介导的降解。尽管 IRP结合共有IRE,研究显示IRP-2 也可以识别一个专有的IRE子集(IRE subset)。
