免疫组织化学

灌注:1)不含Ca2+的台氏液,然后是2)多聚甲醛-苦味酸固定剂和3)10%蔗糖的PBS溶液作为冷冻保护剂。快速解剖组织,并在含有10%蔗糖的0.1 M磷酸盐缓冲液(pH值7.4)中保存过夜。

将载玻片固定的组织切片与封闭缓冲液在室温下培养1小时。将一抗在封闭缓冲液中稀释至适当的工作稀释度。除去封闭缓冲液,然后将载玻片与AB5519在2-8°C下培养18-24小时。在PBS中冲洗3次后,将切片与Cy3偶联的二抗在室温下培养60分钟。固定在含有0.1%对苯二胺的PBS和甘油(1:3)的混合物中后,用Nikon Microphot-SA落射荧光显微镜检查切片。
抗强啡肽原抗体是针对强啡肽原的抗体,可用于IH。
