General description
组蛋白赖氨酸N-甲基转移酶2D(EC 2.1.1.43;UniProt O14686;也称为MLL4,ALL1相关蛋白,赖氨酸N-甲基转移酶2D)由人的KMT2D(也称为MLL4,ALR,KABUK1,MLL2)基因(基因ID:8085)所编码。MLL4使组蛋白H3的Lys-4甲基化,形成H3K4单甲基化和二甲基化(H3K4me1/2),它代表转录增强子的特异性标签。MLL4也是MLL3/4蛋白复合物的一个组分,在细胞分化和动物发育过程中发挥重要作用。MLL4定位于细胞核,并在大多数胚胎和成年组织中表达。MLL4的缺陷(在文献中通常称为MLL2和KMT2D)与歌舞伎综合征和先天性心脏病有关。MLL4/MLL2/KMT2D基因的失活性突变与许多类型的癌症有关,包括髓母细胞瘤,非霍奇金淋巴瘤,弥漫性大B细胞淋巴瘤,前列腺癌,乳腺癌,肝细胞癌,卵巢癌,肺癌,膀胱癌,头&颈癌和胰腺癌。(参考文献:Lee, J.E., et al.
