General description

Tristetraprolin(TTP,Nup 475,TIS11,G0S24或ZFP36)是CCCH串联锌指蛋白家族的成员。是一种作为反式作用因子的核RNA结合蛋白。它参与细胞因子基因的转录后调节,最主要的是肿瘤坏死因子α(TNF-α)。在被脂多糖激活后,TTP在3′-非翻译区(3′-UTR)的富含AU的结构域结合TNF-αmRNA并诱导腺苷酸化,从而使mRNA不稳定并标记其降解。为了产生这种效应,TTP将其他反式作用因子募集到其C末端结构域,并将mRNA衰变酶募集到其N末端结构域。TTP通过促分裂原活化蛋白激酶激活的蛋白激酶2(MK2)和p53驱动的磷酸化而失活。因此,TTP/MK2调节TNF-α的生物合成并介导涉及TNF-α的炎症反应。在小鼠中敲除TTP会产生关节炎,脱发,皮炎,以及与TNF-α过表达相关的一系列其他表型。
观察值〜40 kDa。 Uniprot给出的计算分子量为34 kDa。在某些裂解物中,〜53 kDa处出现未表征的条带。
