General description

GAD65是II类脱羧酶,可借助辅因子5′-磷酸盐催化谷氨酸生物合成GABA。GAD65位于突触前的GABA能端,据报告可提供GABA的局部囊泡池,从而促进瞬时抑制性神经传递,尽管GAD65也可能在更持久的强直抑制信号中起作用。其作为α和β亚基的二聚体存在,并受选择性剪接调节,可产生GAD67亚型。GAD65还经历翻译后修饰,例如N末端半胱氨酸残基的棕榈酰化和多个α-亚基丝氨酸残基的磷酸化,从而实现膜锚定和其他囊泡活性。GAD65与抑制性神经传递失败导致的神经系统疾病有关,例如亨廷顿氏′病和癫痫。除了其CNS的存在外,GAD65还在胰腺β细胞中产生GABA,该区域的功能障碍可能与胰岛素依赖型糖尿病的发病机理有关。
