General description

微管相关蛋白2(MAP-2)是一种丰富的神经元细胞骨架蛋白,可与微管蛋白结合,稳定微管(Herzog and Weber, 1978),并且是神经元的极佳标记物。MAP-2对于神经元形态的发育和维持至关重要(Matus,1991)。已经在神经元中鉴定出MAP-2的三种主要亚型,即高分子量MAP2a和MAP2b,以及低分子量MAP2c(由MAP2基因的交替剪接产生)。(Chung et al.,1996)。低分子量亚型MAP2c在发育中的大脑中表达,并在大脑成熟过程中被下调,而高分子量MAP2b在发育中和成年大脑中均表达。MAP2a仅在大脑成熟后出现(Tucker,1990)。全部这些形式通过羧基末端附近的域与微管结合,该域包含31个氨基酸基序的三个或四个相似重复序列(Lewis et al.,1988)。MAP-2与MAP-4和tau蛋白一起属于与微管相关的热稳定蛋白家族。
