谷氨酸脱羧酶（GAD; E.C.4.1.1.15）是负责将谷氨酸转化为γ-氨基丁酸（GABA）的一种酶，γ-氨基丁酸是大脑较高区域的主要抑制递质，在胰岛中被认为是一种旁分泌激素。GAD的两种分子形式（65 kDa和67 kDa，两种形式之间的氨基酸相似度为64％）是高度保守的，两种形式都在CNS、胰岛细胞、睾丸、输卵管和卵巢中表达。两种亚型在大鼠和小鼠的脑细胞质中分布（Sheikh，1999）。GAD65是一种两亲性膜锚定蛋白（585个氨基酸），由人类10号染色体进行编码，负责囊泡GABA的产生。GAD67位于细胞质中（594个氨基酸），由2号染色体进行编码，其可能是导致大量细胞质GABA产生的原因。GAD的表达在大鼠脊髓神经发育过程中发生变化。GAD65在E13周围的连合轴突中瞬时表达，但在第二天被下调，而GAD67的表达主要在这些神经元的躯干中发生上调（Phelps，1999）。在成熟的大鼠胰腺中，GAD65和GAD67似乎存在差异化定位，GAD65主要存在于含胰岛素的β细胞中，而GAD67存在于含胰高血糖素的（A）细胞中（Li，1995）。GAD67的表达具有特别的可塑性，并且可以响应实验操作（例如神经元刺激或横切）或疾病进展以及诸如精神分裂症等紧急疾病而发生变化（Volk，2000）。两种GAD亚型的共定位也显示出GAD65/GAD67分布的变化，这一变化与某些疾病状态（例如IDDM和SMS）相关。

Biochem/physiol Actions
