General description
LINE-1反转录转座元件ORF1蛋白(UniProt:Q9UN81;也称为L1ORF1p,LINE反转录转座元件1,LINE1反转录转座元件1)由人的LIRE1(也称为LRE1)基因编码。LINE1-ORF 1P属于转座酶22家族。它主要是胞浆蛋白,富含应激颗粒,但在核仁中可能很少被检测到。它包含介导其同三聚化的卷曲螺旋结构域。C末端结构域的RNA识别基序(RRM)是其适当的RNA结合活性所必需的。LINE1-ORF 1P是一种核酸结合蛋白,对于基因组中LINE-1元件的反转录转座至关重要。据报道,它还起结合其自身转录物的核酸伴侣的作用,因此优先动员编码它们的转录物。反转录转座是一个复杂的过程,涉及LINE1的转录,其RNA转运至细胞质,双顺反子RNA的翻译,核糖核蛋白颗粒的形成,其重新导入细胞核,然后在整合位点靶向启动反转录。体细胞LINE1插入在大多数组织中很少见,但在脑、睾丸和许多癌症类型中发现,并与致癌作用有关。在p53突变型肿瘤中始终检测到其表达,但在p53正常的Wilms肿瘤中检测到很少或没有表达。(参考文献:Goodier, JL et al.
