General description
p70s6激酶是一种普遍存在的细胞质蛋白,在细胞因子和生长因子(包括表皮生长因子和成纤维细胞生长因子)的反应中被激活。它位于mTOR/PI3K通路的下游,在多个残基上被磷酸化:Thr389、Thr229、Thr421、Ser371、Ser404、Ser411、Ser418和Ser424。Thr389、Ser371和Ser404位于连接子结构域;而Thr421、Ser411、Ser418、Ser424位于自抑制结构域,Thr229位于催化结构域。这些位点可能按等级顺序被磷酸化,导致p70s6激酶的激活。p70 S6激酶磷酸化40S核糖体蛋白(rpS6)上的S6蛋白,参与5’端寡聚嘧啶束mRNA分子调控、蛋白合成、细胞生长、增殖和细胞存活等多个过程。这种激酶的活性可能在糖尿病、癌症和衰老中起重要作用。
观测分子量~70/85 kDa。 该蛋白可识别α 1和α 2亚型。 在某些裂解物中,根据不同的处理条件,未表征的条带出现在〜18和〜200 kDa处。
