A1 Broth is a powdered microbiological medium designed for the selective detection of fecal coliforms, including Escherichia coli. A1 Broth is formulated with casein enzymic hydrolysate, providing nitrogen, amino acids, and peptides essential for bacterial growth, while lactose and salicin serve as fermentable carbohydrates to support acid and gas production for coliform identification. Sodium chloride maintains osmotic balance, ensuring cell stability, and Triton X-100 acts as a surfactant to inhibit non-target organisms and enhance nutrient dispersion. Together, these components create a selective and nutrient-rich environment ideal for detecting fecal coliforms.

A1 Broth is widely used for detecting fecal coliforms, including Escherichia coli, in water quality monitoring and food safety testing using the Most Probable Number (MPN) method. It is applied in environmental testing of wastewater, seawater, and other samples for coliform contamination, as well as in food and beverage testing to ensure microbiological safety.

Suspend 31.5 g in 1 litre distilled water. Heat to dissolve the medium completely. Distribute 10 ml amounts into tubes containing inverted Durham′s tubes. Sterilize by autoclaving at 121°C for 10 minutes.
