采用过碘酸处理时，乙二醇被氧化成醛。与Schiff′s Reagent（由碱性副品红和偏亚硫酸钠组成的混合物）反应后，碱性副品红化合物释放，将含有乙二醇的胞内组分染为红色。可通过PAS操作步骤染色胞内成分包括糖原、真菌细胞壁、基膜、某些上皮硫粘蛋白和唾液黏蛋白、中性粘液质、甲状腺的胶质材料以及脑垂体的中间部。
