Application
罗丹明123用于:
- 作为P-糖蛋白(P-gp)的底物,可在荧光积累测定中读出功能性转运体活性
- 标记细胞,通过流式细胞术估计P-gp的表达情况
- 在转运研究中,添加至顶端到基底外侧(AB-转运)或基底外侧到顶端(BA-转运)
- 检测线粒体膜电位(ΔΨm)
Biochem/physiol Actions
罗丹明123通常作为官能报告物,报告流式细胞术中的P-糖蛋白(P-gp)。
最常用于流式细胞术的荧光染料,可用作Pgp的功能性报告分子。在癌症治疗中,MDR蛋白的功能测定是比MDR蛋白表达水平更好的预后指标。Rh 123可用于多参数分析而无荧光干扰,可与常用蛋白质标记染料组合使用,如PE-Cy5和AMCA(7-氨基-4-甲基香豆素-3-乙酸)。近期报道表明,Rh123也可能是MRP1的底物。
