General description

β-淀粉样蛋白前体蛋白 (APP) 是由蛋白水解酶β和 γ-分泌酶依次裂解，生成以Aβ1-42和 Aβ1-40型为主的β-淀粉样蛋白 (Aβ) 肽。分泌的Aβ肽通过再吸收机制和之后的内体降解过程降解，或通过胞外胰岛素降解酶降解。Aβ的胞外积累导致聚集体和原纤维的形成，最终形成淀粉样蛋白沉着物神经炎斑，其是阿尔茨海默病 (AD) 的标志物。
β-淀粉样蛋白基因位于人染色体21q21上。
该抗体能够特异性识别β-淀粉样蛋白。该抗体可识别位于β-淀粉样蛋白的氨基酸1-12内的表位。在使用甲酸处理的、福尔马林固定的、石蜡包埋的和Methacarn固定的人阿尔茨海默氏病（AD）脑组织切片中，该抗体能够对皮质内的淀粉样斑块和血管中的淀粉样沉积物进行特异性染色。

Application

小鼠抗ABETA单克隆抗体被用于治疗具有淀粉样蛋白积聚和学习能力缺陷的老年WT PDAPP小鼠，以期改善学习能力并减少积累，但未观察到反应。
抗β-淀粉样蛋白单克隆抗体已被用于免疫沉淀分析、微阵列免疫测定和微阵列。
该抗体可用于免疫组织化学、免疫印迹、ELISA和竞争性ELISA。此外，该抗体已被用于中和表达突变型淀粉样蛋白前体蛋白的转基因小鼠脑中的Aβ组装体，以及利用近红外光谱的体内深部组织成像。

Biochem/physiol Actions

β-淀粉样蛋白基因是胰岛素降解酶（IDE）的底物。它在阿尔茨海默病（AD）和2型糖尿病（DM2）的发病机制中发挥主要作用。
