General description
α-辅肌动蛋白是肌肉以及非肌肉细胞中的一种100kD肌动蛋白结合蛋白。在平滑肌中,组织的密集体和斑块中存在α-辅肌动蛋白,而它与定义了正常骨骼肌中肌肉肉瘤的Z盘有关。单克隆抗-α-辅肌动蛋白抗体可用于免疫印迹,以研究α-辅肌动蛋白的膜锚定位点和免疫化学鉴定。它也可以用于免疫荧光研究α-辅肌动蛋白在培养的细胞和组织中的定位。鼠抗α辅肌动蛋白抗体与鸡成纤维细胞发生特异性反应。该产品还显示出对小鼠和人类细胞的反应性。
编码α-辅肌动蛋白的基因定位于人类染色体11q13.2。α-辅肌动蛋白属于血影蛋白(spectrin)超家族。

Application
单克隆抗α-肌动蛋白抗体(稀释1:500) 可用于天然蛋白质结合测定。它还可用于蛋白质印迹、微阵列、免疫组化和免疫荧光显微镜。

采用单克隆小鼠抗-α肌动蛋白(克隆 EA-53),以1:1000的稀释度对新生和成年小鼠心脏的心肌细胞进行免疫荧光分析。
Biochem/physiol Actions
α-肌动蛋白调节肌肉收缩和细胞骨架组织。它促进细胞迁移并用作肌动蛋白交联剂。在异质性肥厚性心肌病和青少年起病性房颤患者中,已发现α-辅肌动蛋白基因发生突变。
