Biochem/physiol Actions

半胱天冬酶3是细胞凋亡的主要效应因子。在凋亡通路中，它在半胱天冬酶9的下游起作用。凋亡期间，经由蛋白水解活化后，变为17-19 kDa(p17, p18)和12 kDa(p12)活性亚基。小鼠CASP-3的任意变化都会导致过早死亡。已发现，基因缺失会影响大脑发育，导致增生和细胞部署混乱。临床上与亨廷顿病(HD)有关。半胱天冬酶3可激活核酸内切酶CAD(caspase activated DNAse I)降解染色体DNA，导致细胞死亡。

General description

半胱天冬酶(Caspase)是介导细胞死亡的胞内蛋白酶家族，是细胞凋亡的主要效应器。目前已发现14种半胱天冬酶，根据底物特异性可分为3个亚家族。半胱天冬酶3又名CPP32、Yama、apopain。半胱天冬酶3以无活性的32 kDa酶原形式存在于细胞质中。
半胱氨酸蛋白酶-3 (CASP3)属于胱氨酸蛋白酶类。CASP3是一种半胱天冬酶，在半胱氨酸蛋白酶8抗原和半胱氨酸蛋白酶9抗原引发剂半胱天冬酶的帮助下被裂解和激活。CASP3定位于人染色体4q35.1。

与结合到KLH的人胱天蛋白酶3的切割位点相对应的合成肽。

供应0.01 Mp磷酸盐缓冲盐水，pH 7.4，含15 mM叠氮化钠。

Preparation Note

储存温度–20℃。为长期使用，产品可以储存，产品可以在2-8℃下储存长达一个月。对于长期储存，在-20℃冷冻工作等分试样不建议重复冷冻和解冻，或储存在无霜冰箱中。如果长期储存时出现轻微混浊，使用前通过离心澄清溶液。如果在12小时内未使用，工作稀释液应丢弃。
