Biochem/physiol Actions

单克隆抗-磷酸化组蛋白H3（pSer28）抗体可与近N末端的Ser28上发生了磷酸化的组蛋白H3发生特异性反应，但不会检测未磷酸化的表位。它可在有丝分裂开始时检测组蛋白分子（前期、中期，后期初期较弱），后期末期则不会检测。目前已在人、牛、仓鼠和小鼠中观察到反应性。
有丝分裂刺激、致癌转化或致癌 ras 表达的诱导都伴随着 H3 N-末端结构域丝氨酸-10 磷酸化的增加。研究表明，磷酸化的 H3 与受刺激细胞中的 c-fos 和 c-myc 基因相关。H3在丝氨酸-10和-28的磷酸化与有丝分裂染色体凝聚的诱导相一致。
H3磷酸化可能通过调节染色质结构和释放延伸阻断来促进原癌基因诱导。在有丝分裂后，H3迅速去磷酸化，且丝氨酸-10/28 reo在间期的剩余时间内主要是未磷酸化的。PP1已被鉴定为H3磷酸酶。
该抗体不会检测未磷酸化的表位。它可在有丝分裂开始时检测组蛋白分子（前期、中期，后期初期较弱，后期末期不检测）。

General description

组蛋白H1和组蛋白-3(H3)的N端尾部在染色质纤维的折叠和相互关联中起关键作用。目前已知的两个磷酸化位点位于 H3 的 N-端；丝氨酸-10 和丝氨酸-28。哺乳动物主要包括三类组蛋白H3变体：复制组蛋白（H3.1 和 H3.2）、替换组蛋白 (H3.3) 和着丝粒组蛋白 (Cenp-A)。组蛋白H3位于人类染色体 6p22 上。

对应于人组蛋白H3氨基酸的合成肽。

Physical form

含有1% BSA及15 mM叠氮化钠的 0.01 M磷酸盐缓冲液（pH 7.4）溶液。

Preparation Note

从在生物反应器中生长的杂交瘤细胞培养上清中纯化所得
如需持续使用，可在2至8 °C下储存一个月。如需更长期储存，可分装后冷冻溶液。不建议反复冻融。不建议储存在“无霜”冰箱中。如果在长期储存后出现轻微混浊，可在使用前通过离心使其澄清。工作稀释样品在稀释小时之后应被丢弃。
