General description
LNX1 (ligand of numb-protein X 1) is the PDZ (PSD95, Dlg1, and zo-1) domain-containing member of the RING (really interesting new gene) finger-type E3 ubiquitin ligase family. It participates in tumorigenesis. It is associated with various signal transduction pathways such as notch pathway. It consists of different protein-protein interaction domains including a RING finger and four PDZ domains.

Application

Biochem/physiol Actions
LNX1 (ligand of numb-protein X 1) interacts with the phosphotyrosine-binding (PTB) domain which is essential for NUMB functions i.e. cell cycle control. It helps to cluster PTB-containing proteins along with functionally related transmembrane proteins in specific membrane compartments. In addition, it has been shown that downregulation of LNX1 causes cell cycle arrest in G0/G1 phase by suppression of of β-catenin, MAPK (mitogen activated protein kinase), NF (nuclear factor) κB, c-Myc-dependent pathway and by activation of p53, TGF (transforming growth factor)-β-dependent pathway.
