General description

The gene NAA25 (N(α)-acetyltransferase 25) is mapped to human chromosome 12. The encoded protein is an auxiliary subunit of the NatB (N-terminal acetyltransferase) complex.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Biochem/physiol Actions

The NatB (N-terminal acetyltransferase) complex is responsible for the acetylation of N-terminal methionines in proteins during de novo protein synthesis. It is also responsible for the N-terminal acetylation of tropomyosin, thereby stabilizing and regulating actin remodeling. NAA25 (N(α)-acetyltransferase 25) is required for actin remodeling and cell motility. It regulates Rictor and controls the activity of mTORC2 (mammalian target of rapamycin complex 2). These events modulates Akt (protein kinase B), PKCα (protein kinase C α), and FoxO1 (forkhead box protein O1) activities, thereby influencing cellular homeostasis, cell motility and metabolic events. NAA25 suppresses the induction of autophagy via Akt phosphorylation, thereby stimulating polyQ (polyglutamine) aggregation. Polymorphism in the NAA25 gene region is associated with juvenile idiopathic arthritis.
