角蛋白。角蛋白酶也被用于朊病毒和朊病毒样蛋白的降解。
角蛋白酶 也是金属蛋白酶，存在于细菌和真核生物中。从地衣芽孢杆菌（ Bacilus licheniformis ）培养物中分离的角蛋白酶是单体酶。
角蛋白酶是一种非特异性丝氨酸蛋白酶，能切割非末端肽键。这种酶在 37°-70°C 之间有活性。通常在 70°C 时活性最高。
角蛋白酶活化 0.10% SDS、1.0%CTAB 和 EDTA。角蛋白酶可被吐温 ® 20、DMSO、异丙醇、甲醇和乙醇部分抑制。
应用
角蛋白酶可对原体（EB）、GAG分子和细胞进行酶处理，用于研究 肺炎衣原体 菌株侵入宿主细胞过程中糖胺聚糖（GAG）的作用。
生化/生理作用
角蛋白酶是一类特殊的胞外蛋白水解酶，能降解不溶性角蛋白底物。 在污水处理过程中，对水解污水系统中的毛发、羽毛、胶原蛋白有重要作用。在食品工业、动物饲料配制等方面也有应用。来自家禽工业的不溶性羽毛角蛋白可通过酶水解转化为胶水、饲料、肥料、薄膜或用于生产稀有氨基酸丝氨酸、半胱氨酸和脯氨酸。
制备说明
该酶可溶于无菌水或磷酸盐缓冲液中，浓度为 0.5-1.0 mg/mL。新鲜制备的溶液的活性最好。但是，单次使用的角蛋白酶溶液等分试样可在-20℃ 下储存。
其他说明
在 37℃、pH 7.5 时， 1 个单位的酶每分钟水解酪蛋白，产生的吸光度值相当于 1 μmol (181 μg) 的酪氨酸 的Folin-Ciocalteau 试剂。
这种酶含有一个 C 端 6-组氨酸标签。
