一般描述

本品为绿脓杆菌血清型10提取的 TCA。来源菌株为 ATCC 27316。

应用

脂多糖 (LPS) 是革兰氏阴性菌细胞壁的特征性成分。LPS 及其脂质 A 部分通过 Toll 样受体 4 (TLR4) 刺激先天免疫系统的细胞,TLR4 是 Toll 样受体蛋白家族的成员之一,可识别常见的病原体相关分子模式 (PAMP)。

生化/生理作用

脂多糖(LPS)位于膜的外层,并且在非包封的菌株中,其暴露于细胞表面。它们有助于外膜保持完整,并可保护细胞免受胆汁盐和亲脂性抗生素的影响。

制备说明

本品可溶于水(5 mg/mL)或细胞培养基(1 mg/mL),产生淡黄色浑浊溶液。涡旋并加热至 70-80 o C 后,在盐水中获得了更浓缩但仍浑浊的溶液(20 mg/mL)。脂多糖是在每种溶剂中形成胶束的分子。在水和磷酸盐缓冲盐水中观察到浑浊溶液。有机溶剂不能提供更清晰的溶液。甲醇产生浑浊、带漂浮物的混悬液,而水产生均匀的浑浊溶液。
