General description
MacConkey琼脂用于乳糖发酵和非乳糖发酵肠道细菌的分离、鉴定和计数。此培养基受牛奶和乳制品及药物制剂标准检验方法认可。培养基中的蛋白胨提供含氮和含碳化合物、长链氨基酸、维生素和其他必需的生长因子。乳糖是一种可发酵糖，当培养基的 pH 值低于 6.8 时，会导致酸形成和指示剂颜色变化。氯化钠可维持渗透压平衡。该培养基的选择性作用源自结晶紫和胆盐，它们对大多数革兰氏阳性菌具有抑制作用。革兰氏阴性菌通常在培养基上生长良好，并可通过乳糖发酵能力进行区分。乳糖发酵菌株形成红色或粉红色菌落，并被酸沉淀胆汁区域包围。非乳糖发酵菌株（例如志贺氏菌和沙门氏菌）形成无色透明菌落，通常不会改变培养基的外观。

Biochem/physiol Actions
麦康凯琼脂培养基被视为一种选择性和鉴别性培养基。它有助于区分乳糖发酵菌和非乳糖发酵菌。因此，可用于分离革兰氏阴性细菌。原代平板培养基用于区分大肠杆菌群的成员。

Preparation Note
